Orthogonal experimental design optimization of the alcohol extracting technology of Lithospermum: High peaks are thoroughly separated, but without altering the retention time, thus utilizing the mobile phase elution. There have been numerous reports on this topic. The high concentration ethanol refluxing method is commonly used for the extraction of effective shikonin components. This article also adopts reflux extraction as the orthogonal test method. From the results, the shikonin content in the three reflux extraction samples is very low, and other ingredients suffer severe destruction, which is also proven by the HPLC charts. This proves that the reflux method for extracting Arnebia medicinal materials is debatable, and hopes to draw the attention of peers. According to the comparison analysis of HPLC fingerprints of samples extracted at different temperatures, it is proven that under the condition of 65°C temperature, soaking extraction results in less component loss while achieving higher extraction efficiency and the purpose of saving energy. The test shows that the screened process conditions are stable and feasible, suitable for industrialized production.
